Sex difference in diabetes
In humans, the incidence of Type 2 Diabetes Mellitus (T2DM) has a clear sexual dimorphism, a phenotype recapitulated by many rodent T2DM models. Nevertheless, the main reason has been proposed to be the sex difference in insulin sensitivity. Previously we demonstrated that deletion of α2δ-1, an auxiliary subunit of the high voltage gated calcium channel (HVCC), leads to diabetes in mice in a sex-specific manner despite a similar insulin sensitivity. In my project, we aim to investigate the molecular mechanisms that lead to a higher insulin release in females compared to males.
Methods Primary pancreatic β-cell culture, Electrophysiology (current and voltage clamp), Capacitance measurements, Calcium imaging, Mouse handling and breeding, Dynamic hormone release, MEA.
